In particular, peptides containing <em>N<\/em>-alkylamines often exhibit low reactivity due to their substantial steric hindrance and are prone to problems such as epimerization and reduced yields, making their efficient connection a longstanding challenge. Through extensive investigation, the presenters developed two distinct activation methods and successfully achieved the rapid connection of highly sterically hindered peptides under mild conditions by precisely controlling the electrophilicity of the peptide C-terminus and the pH of the reaction system, while effectively suppressing epimer formation. The methodology is also applicable to substrates containing <em>N<\/em>-alkylamines, which have traditionally been difficult to employ in peptide ligation, and is therefore highly regarded as an excellent approach for the efficient synthesis of <em>N<\/em>-alkyl peptides. In recent years, convergent synthetic strategies have become increasingly important for improving the efficiency of peptide synthesis. However, reports of practical implementation remain limited. In this presentation, a continuous flow process for a pharmaceutical intermediate was examined. The study demonstrated an integrated development approach that covers both laboratory studies and manufacturing, and it provided a clear pathway toward practical implementation. In particular, complex reaction parameters were efficiently and rationally optimized using definitive screening design (DSD). Advanced control and monitoring systems were implemented, enabling robust and stable continuous flow production. This achievement is highly noteworthy. Furthermore, practical insights into challenges encountered during manufacturing, including heat removal and line clogging, were disclosed. These findings contributed to the advancement of flow chemistry. In addition, 50 kg-scale GMP manufacturing was successfully completed. The strategy is highly commendable for its rational design and strong scale-up orientation, including the introduction of chirality from inexpensive starting materials, the formation of the 12-membered macro-lactone via RCM, and a convergent synthetic approach employing the Mizoroki\u2013Heck reaction. Conventional methods for amide bond formation that rely on coupling reagents suffer from a number of challenges from the viewpoint of process chemistry. In this work, the presenters developed a dehydrative amidation reaction catalyzed by a novel diboronic acid anhydride, providing a clear solution to these issues. The introduction of a carbazole framework as the core catalyst structure is particularly noteworthy, as it substantially expands the substrate scope compared with existing diboronic acid catalysts. Using this methodology, the catalytic dehydrative amidation of less-reactive aromatic carboxylic acids with amines proceeds under operationally simple conditions without the need for stoichiometric amounts of dehydrating reagents, while generating water as the sole byproduct.
